Systematic Reviews and Meta- and Pooled Analyses Hemochromatosis Gene (HFE) Polymorphisms and Risk of Type 2 Diabetes Mellitus: A Meta-Analysis
نویسندگان
چکیده
The hemochromatosis gene (HFE) has been involved in the etiology of type 2 diabetes mellitus and investigated in numerous epidemiologic studies. The current meta-analysis was conducted to evaluate the gene-disease association in relevant studies. Electronic literature search was performed on June 18, 2011, from databases of PubMed/MEDLINE, EMBASE, and HuGE Navigator. Articles were inspected by 2 authors independently, and data were extracted by identical extraction form. A total of 5,528 type 2 diabetes cases and 6,920 controls in relation to HFE polymorphisms (a cysteine to tyrosine substitution at amino acid position 282 (C282Y) and a histidine to aspartate substitution at amino acid position 63 (H63D)) were included in the meta-analysis (1997–2011). A fixedor random-effect model was used to calculate the pooled odds ratios based on the results from the heterogeneity tests. An increased odds ratio for type 2 diabetes mellitus was observed in persons carrying a D allele at the H63D polymorphism compared with those with an H allele (odds ratio (OR) = 1.21, 95% confidence interval (CI): 1.03, 1.41; P = 0.02). Moreover, carriers of a D allele had a modestly increased risk compared with persons with the wild genotype (OR = 1.12, 95% CI: 1.00, 1.25; P = 0.04). The C282Y variant was not significantly associated with diabetes risk. In summary, persons with a D allele may have a moderately increased risk of type 2 diabetes mellitus.

Background and Aim: Several investigations have evaluated the association of miR-146a rs2910164 and miR-27a rs895819 single-nucleotide polymorphisms with susceptibility to type 2 diabetes (T2D). However, the findings are conflicting and inconclusive. Therefore, this meta-analysis performed to investigate the association between these polymorphisms and T2D risk. Methods: Studies were identifie...

Objective(s): Diabetes mellitus (DM) is a major worldwide public health challenge, for which gene therapy offers a potential therapeutic approach. To date, no systematic review or meta-analysis has been published in this area, so we examined all relevant published studies on rodents to elucidate the overall effects of gene therapy on bodyweight, intraperitoneal glucose...
متن کاملRe: "hemochromatosis gene (HFE) polymorphisms and risk of type 2 diabetes mellitus: a meta-analysis".
We read with great interest the article by Rong et al. (1) on the associations of hemochromatosis gene (HFE) polymorphisms (C282Y and H63D) with type 2 diabetes mellitus. The authors stated, “... persons with a D allele may have a moderately increased risk of type 2 diabetes mellitus” (1, p. 461) (additive model (D allele vs. H allele): odds ratio (OR) = 1.20, 95% confidence interval (CI): 1.03...

Background: Many studies have reported the association of estrogen receptor α gene (ESRα) ESRα PvuII T>C, XbaI A>G and BtgI G>A polymorphisms with Knee osteoarthritis (KOA) risk, but the results remained controversial. In order to drive a more precise estimation, the present systematic review and meta-analysis was performed to investigate the association between ESRα polymorphisms and KOA susce...

The association between Guanine Nucleotide Binding protein β protein polypeptide 3(GNB3) C825T polymorphism and obesity has recently been reported. However, the findings remain inconclusive. The aim of this systematic review and meta-analysis was to detect the relationship between GNB3 C825T polymorphism and obesity.
